Immunohistochemistry on snap frozen or paraffin embedded tissue.
Suggested working dilution for fresh rat tissue is 1:400 and for fresh human tissue is 1:20. Fixation: acetone 5 min at 4°C, paraformaldehyde, paraffin. Tissue pretreatment: microwave treatment 2 x 5 min at 750 W for paraffin tissues.

General description
Surfactant protein D is a Ca2+ dependent carbohydrate binding protein with a structure similar to other C-type mammalian lectins. Its molecular weight is 620 kDa under non-reducing conditions and 43 kDa under reducing conditions. The function of SP-D is not clearly defined. SP-D enhances the production of oxygen radicals in alveolar macrophages and regulates some actions of SP-A.

Specificity
MAB3132 recognizes an epitope localized in the lectin domain of rat surfactant protein D (SP-D). Reacts with alveolar type II cells, alveolar macrophages, types of Clara cells (in which SpD is highly concentrated), SP-A+ and SP-A-, and extracellular deposits. Alveolar type III cells are negative. No reaction on rat skin, colon, kidney or liver. SP-A cross reacitivity is typically evident at very low dilutions (1 to less than a few hundred) by ELISA.
Storage and Stability
Maintain lyophilized material at 2-8°C for up to 12 months from date of receipt. Reconstitute lyophilized material with 500 µL of sterile distilled water. After reconstitution, maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.
